PRESENT: Probe International Inc. is now diversifying and focusing on our earlier technology (4,315,906 Cold insoluble globulin, its purification and use) which has been further developed resulting in methods for producing low endotoxin (< 10 EU/mg) fibronectins from human and various other mammals.
Human fibronectin is purified from single blood donors from FDA approved USA blood centers. All units are tested and are negative for antibodies to HIV, HCV, HTLV, HBc and nonreactive for HBsAg, STS, HCV RNA, HIV RNA and WNV RNA by FDA required testing.
Fibronectin is purified employing ligand specific affinity chromatography with proprietary additives which reversibly bind and protect fibronectin bioactive domains during purification. This prevents loss of bioactivity and denaturation of the fibronectin. Fibronectin purified by this improved bioactive site protecting technology results in the retention of native fibronectin bioactivity in our purified, low endotoxin products..
Probe International Inc. manufactures purified fibronectins which promote cell adhesion, enhancement of anchorage dependant and independent primary and secondary malignant cell attachment and growth. Fibronectin extra domain A following interaction with TLR4 signals tissue injuries in the absence of infection and the need for tissue repair, thus providing a new tool for investigating tissue repair processes in vitro. Fibronectin is also employed as a non-immune opsonization for S. aureus, S. pyogenes and various other pathogens providing research avenues for investigating innate host defense mechanisms.
Fibronectin coated tissue cultures are proving useful in Stem cell research. Fibronectin is used as a coating agent for coverslips, microtiter wells and plates and various other tissue culture vessels to encourage Stem cell attachment and growth. Fibronectin promotes Stem cell attachment, growth and yield of neural cells during the final stages of mouse embryonic stem cell (mESC) in vitro differentiation into neural cell types. Mesenchymal stem cell migration is regulated by fibronectin through α5β1-integrin-mediated activation of PDGFR-β and potentiation of growth factor signals.
Fibronectin provides a functionally defined and effective feeder-free surface for the attachment and maintenance of embryonic stem cells in a pluripotent state. It is an extracellular matrix protein that is found abundantly in blood, connective tissues, and remodeled matrices also associated with the epithelial to mesenchymal transition of migratory cells - including tumor cells with stem cell-like properties. Fibronectin performs essential functions in collagen fibrillogenesis, and as either a general cell adhesion molecule or as a modulator in binding between cell surfaces and the extracellular matrix. Fibronectin matrix assembly is essential for normal vertebrate development, and apparently contributes to the generation of tumor metastases by supporting the establishment and persistence of premetastaticniches. Fibronectin is secreted as a disulfide-linked dimer of 230-270 kDa, comprised of three types of repeating modules that mediate interactions with extracellular matrix components (including fibronectin itself), and cells via integrins and other fibronectin receptors.
Microbial pathogenesis is influenced by fibronectin binding adhesins present in several strains of Streptococcus and Staphylococcus. Our purified human and species-specific fibronectin offers investigators a probe to further understand the role of pathogen binding adhesion interaction with host extracellular matrix proteins. Elucidation of such mechanisms may lead to discovery of new modes of treatment for the highly virulent multi drug resistant microbial strains emerging through out the world.
